Cyanine 5 Tyramide is widely used in fluorescence-based detection methods, particularly in tyramide signal amplification (TSA) techniques. It enables highly sensitive visualization of low-abundance targets in immunohistochemistry (IHC), immunocytochemistry (ICC), and in situ hybridization (ISH). Upon activation by horseradish peroxidase (HRP) in the presence of hydrogen peroxide, the tyramide moiety forms reactive radicals that covalently bind to tyrosine residues on nearby proteins, depositing the Cy5 fluorophore precisely at the target site. This localized deposition allows for significant signal amplification with minimal background, making it ideal for multiplex labeling when combined with other fluorophores. Its bright far-red fluorescence is well-suited for imaging in thick or autofluorescent tissues and is compatible with standard Cy5 filter sets and laser systems.
