Casegravol is primarily used in the treatment of nausea and vomiting associated with chemotherapy, particularly in cancer patients undergoing highly emetogenic treatments. It functions as a neurokinin-1 (NK1) receptor antagonist, helping to block substance P in the central nervous system, which plays a key role in the vomiting reflex. It is often administered in combination with other antiemetics such as 5-HT3 receptor antagonists and corticosteroids to provide enhanced protection against both acute and delayed chemotherapy-induced nausea and vomiting (CINV). Due to its long half-life and strong binding affinity, it offers prolonged symptom control, improving patient comfort and compliance during intensive cancer therapy. Casegravol is also being studied for use in postoperative nausea and vomiting, as well as in conditions involving central emesis pathways.
