3',4',5,7-Tetramethoxyflavone (TML) is a potent anti-inflammatory polymethoxyflavone that specifically targets mast cells and keratinocyte inflammatory pathways, making it ideal for sensitive, reactive, and inflamed skin conditions.

Scientific research demonstrates TML's powerful mast cell stabilization properties. It inhibits human mast cell degranulation and blocks the release of inflammatory mediators including histamine, β-hexosaminidase, TNF, and CCL2. This occurs through reduction of intracellular calcium and NF-κB activity. In keratinocytes, TML suppresses TNF-α-induced inflammatory cytokines (IL-6, IL-8/CXCL8, VEGF) via mTOR pathway inhibition, directly addressing the inflammatory cascade in conditions like atopic dermatitis, psoriasis, and chronic urticaria.

Clinical testing shows excellent tolerability. An open-label pilot study with a TML-containing lotion demonstrated no irritation in 25 highly sensitive-skin patients. In a small case series (n=8) including atopic dermatitis and psoriasis patients, lesions showed improvement after 4 weeks of twice-daily application. As part of the polymethoxyflavone (PMF) class, TML shares anti-photoaging benefits demonstrated by related compounds including enhanced collagen I production, increased hyaluronic acid, and reduced oxidative stress in UV-exposed skin.
