Used in RNA oligonucleotide synthesis as a protected phosphoramidite building block. The 5'-DMT group provides temporary protection for the 5'-hydroxyl during solid-phase synthesis, enabling stepwise assembly of RNA strands with high efficiency and specificity. The benzoyl (Bz) group protects the exocyclic amine on the adenine base, minimizing side reactions during coupling and oxidation steps. After synthesis, the DMT group is removed under mild acidic conditions to allow for chain elongation or final deprotection, while the benzoyl group is cleaved during standard ammonia-based deprotection. Commonly used in the production of antisense oligonucleotides, siRNA, and other functional RNA molecules for research, diagnostics, and therapeutic development.
