According to the research studies presented, Palmitoyl Pentapeptide-4 (also known as Pal-KTTKS) has shown efficacy in reducing wrinkles and improving skin appearance. Here are the key findings summarizing its efficacy:
1. In a 12-week, double-blind, placebo-controlled, split-face study on 93 women aged 35-55:
- Quantitative image analysis showed a significant reduction in fine lines/wrinkles with 3 ppm Pal-KTTKS compared to placebo, with a reduction of around 10-15% from baseline at weeks 8 and 12 (exact numbers: 98.05 at baseline, 74.86 at week 12 for Pal-KTTKS vs 101.8 at baseline, 101.25 at week 12 for placebo).
- Expert graders also identified significant improvement in fine lines/wrinkles with Pal-KTTKS at weeks 8 and 12 compared to placebo (grading scale +4 to -4, Pal-KTTKS scored around 0.6-0.8 higher than placebo).
- Skin texture was graded as significantly improved by expert graders with Pal-KTTKS at weeks 4 and 8 compared to placebo.
2. In an 8-week split-face study on 15 women aged 35-65:
- Visioscan measurements showed a significant reduction in wrinkle depth for both crow's feet (98.05 at baseline to 74.86 at week 8 for Pal-KTTKS) and under-eye wrinkles (119.96 at baseline to 85.82 at week 8) with 7% Pal-KTTKS compared to placebo.
- Cutometer measurements showed a significant increase in skin elasticity for both crow's feet (0.6855 at baseline to 0.8218 at week 8) and under-eye (0.7421 at baseline to 0.8341 at week 8) with 7% Pal-KTTKS compared to placebo.
- TEWL (skin barrier function) significantly improved with 7% Pal-KTTKS (e.g. crow's feet 13.85 at baseline to 5.88 at week 8) compared to placebo.
- Rao-Goldman wrinkle scores reduced significantly from baseline to week 8 with 7% Pal-KTTKS (e.g. crow's feet 3.73 to 2.47) but not with placebo.
- Participants reported significantly higher satisfaction scores with 7% Pal-KTTKS compared to placebo for reducing wrinkles.
The studies demonstrate that Palmitoyl Pentapeptide-4, even at low concentrations like 3-7 ppm/%, can provide significant improvement in wrinkle reduction, increased skin elasticity and improved skin barrier function over 8-12 weeks of use, without any irritation or side effects.
